Understanding Empliciti®

Empliciti® (elotuzumab) is a prescription medicine indicated, in specific treatment settings and regimens, for the treatment of adult patients with multiple myeloma:

  • In combination with lenalidomide and dexamethasone, for patients who have received one to three prior therapies.
  • In combination with pomalidomide and dexamethasone, for patients who have received at least two prior therapies including lenalidomide and a proteasome inhibitor.

Empliciti® is a SLAMF7-directed immunostimulatory antibody administered by intravenous infusion. Elotuzumab is a humanized IgG1 monoclonal antibody that specifically targets the SLAMF7 (Signaling Lymphocytic Activation Molecule Family member 7) protein.

How Empliciti® Works:

  • Targets the SLAMF7 protein: Empliciti® contains elotuzumab, a humanized IgG1 monoclonal antibody that specifically targets SLAMF7.
  • Targets SLAMF7 on myeloma cells: SLAMF7 is expressed on myeloma cells independent of cytogenetic abnormalities. SLAMF7 is also expressed on natural killer (NK) cells, plasma cells, and at lower levels on specific immune cell subsets of differentiated cells within the hematopoietic lineage.
  • Directly activates NK cells: Elotuzumab directly activates NK cells through the SLAMF7 pathway.
  • Mediates killing of myeloma cells: Elotuzumab also targets SLAMF7 on myeloma cells and interacts with Fc receptors on effector cells to mediate the killing of myeloma cells through antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP).

FDA Approval:

Empliciti® (elotuzumab) received its initial U.S. FDA approval in 2015. Empliciti® is FDA approved for use in combination with lenalidomide and dexamethasone, and in combination with pomalidomide and dexamethasone, for the multiple myeloma settings described above.
